defining parameters

hey,

I do simple time lapse experiments with e.coli 100x lenses.
length of e.coli is 5 microns by my camera scale.
but the software doesn't recognize any cells, I would love help redefining the parameters if needed, couldn't find an easy explanation for not programmers :)
