I suspect your use case would be better addressed with a different pattern. For example, interior mutability should be avoided at all costs, since it breaks the DAG.

If you really must refresh a cell manually, consider using a run button or refresh button component. I use this pattern a ton:

# Cell 1: run button will kick off manual refresh

button = mo.ui.run_button(label="Refresh Singleton")
button

# Force cell refresh when button is clicked 
button.value
my_singleton = MySingleton()

# Expose refresh button wherever
button

I'm not sure if there's a component that binds to keyboard shortcuts. Could be a new feature request?

"Re-run all cells" doesn't restart the kernel; it re-runs cells in the same process, so module state stays.

Restart isn't in the keymap (hotkeys.ts), so it can't get a shortcut, but the command palette has it: Cmd/Ctrl-K, type restart, Enter, then confirm.

To have the restarted kernel run everything, turn on "Autorun on startup" (Settings > Runtime, off by default in 0.25.1), or for one notebook:
