In playing with venom arrows, I've noticed that they increase venom buildup over time, and only two arrows are necessary to drop a megasloth.
I figure venom ought to be related to body size, and definitely made weaker.
As it stands, one can shoot a few arrows, hide behind cover and watch enemies fall down after some time.

What amount of arrows should drop a megasloth, though?

Say, you fire twenty arrows and wait a while, they die, is that acceptable? Should the venom effect only be applied if the arrow penetrates? Should venom arrows cost more to make and use?

This should already be the case. The problem is the venom arrows are using the same hediff from the Cobra bite. The values were calibrated specifically around said Cobra bites and likely don't scale correctly when applied to projectiles.

Also may be worth investigating actual Cobras, e.g. put Cobra vs Megatherium, just to rule out bugs in the venom system.

Some interesting suggestions:

  • Make poisonous ammo require poison resource to craft
  • Poison can be gathered from certain poisonous animals, such as cobras
  • Alternatively, poisonous plants can be planted for purpose of eventual concentration into poison (perhaps using a campfire)
  • Small doses of poison is often therapeutic, so it could be used as a requirement in some drugs

Suggestions:

  • Snakes are milkable for (one kind of) poison
  • Curare or similar plants with nerve agent effects can be grown (new plant) and processed (at a campfire at a low rate, or at a stove at a higher rate) into poison
  • Poison arrows require poison rather than healroot

Snakes are obviously already terrible to keep around, perhaps they could have high chances to bite on nuzzling or something, so your colony has a hard time handling them.

Poisonous plants could slowly apply all sorts of nerve damage on its harvesters.

And even more:

  • Antidotes could be added, either as store-bought resources or somehow through research/recipe chains
  • Carrying an antidote on a pawn causes them to immediately use it once they got a lethal dose of poison
